一个包含小鼠Il7基因座及周边序列（96kb的5'端和17kb的3'端）的BAC克隆（RPCI-295A3）在大肠杆菌中通过同源重组进行了修改，引入了cre重组酶的cDNA和一个由FRT序列标记的Zeo抗性基因，位置在Il7 mRNA的第1个外显子1的起始位置，这会替换掉整个外显子1的开放阅读框以及内含子1的第1个核苷酸。重组的BAC被转化到了表达FLP的E. coli菌株FLP-294中，以去除Zeo抗性基因。线性化的BAC通过卵母细胞的核移植技术注入。建立了两条系，其中C线（含有3个转录本）被进一步研究。（来源：J:149246）
