一个包含小鼠Il7基因座及周边序列（96kb的5'端和17kb的3'端）的BAC克隆（RPCI-295A3）在大肠杆菌中通过同源重组进行了修改，加入了人类的IL2RA（CD25）基因和一个由FRT序列引导的Zeo抗性基因，这个基因位于Il7 mRNA的exon1的起始位置，取代了exon1的整个开放阅读框以及intron1的首纳位。重组的BAC被转化到了表达FLP的E. coli菌株FLP-294中，以消除Zeo抗性基因。线性化的BAC通过卵母细胞的核内注射技术导入。建立了两条系，其中B系（携带单个转录本）被进一步研究。（来源：J:149246）
