在5' UTR的上游(ATG起始密码子前50bp)引入了loxP位点，接着在3' UTR的下游(停用密码子后300bp)放置了三个SV40早期终止子重复，同时包含人类胎盘碱性磷酸酶(HPAP)基因和FRT标记的诺卡因抗性基因。将这种靶向载体导入了129系胚胎干细胞。随后，通过生殖细胞内的FLP重组酶，删除了诺卡因抗性基因 cassette。(来源：J:177259)
