在PC3 ES细胞中，通过同源重组将一个IREScreERT2 cassette紧接在Lmx1b基因的3'UTR区域后，接着插入了一个由loxP标记的PGK-neo cassette。随后，来自雄性嵌合体小鼠的Cre介导的重组会删除PGK-neo cassette。通过与C57BL/6小鼠杂交，从敲入等位基因中分离出了由PC3细胞提供的protamine-cre转基因。(来源：J:149241)
