TgNL2转基因是通过修改了"小鼠thy1.2基因"的调控区域（包括从启动子延伸到第4外显子之后的内含子，不包括第3外显子及其前后内含子）来设计的，其上游连接了一个带血小板糖蛋白（Hemagglutinin）标记的猪神经连接素2（Nlgn2或NL2）的cDNA。转基因的始祖鼠经过杂交，与C57BL/6鼠交配后，后代中TgNL2.6品系（来源于这一创始人线）的NL2表达大约增加了1.6倍，并表现出中等程度的表型严重性，相关信息参考文献为(J:137376)。
