这个靶向构建是为了在Egr3基因的3' UTR插入一个IRES-Cre-pA元件。将靶向载体通过电穿孔注入W9.5 ES细胞中。通过同源重组筛选克隆，阳性克隆再注入C57BL/6囊胚中，产生嵌合体供原代。这个等位基因保留了内源Egr3基因的表达，并在Egr3启动子的调控下表达Cre。(来源：J:147957)
