对于携带有Tcrbtm1Slek这一等位基因，且dbeta2-Jbeta2缺失的ES细胞，进行了第二次定向操作。预组装的dbeta1Jbeta1.1复合体被引入，它不包含N和P碱基，与基因组序列相比缺少一个C碱基。这个复合体取代了Dbeta1和Jbeta1.1的基因座。在Jbeta1.2和Jbeta1.3之间插入了floxed的诺卡因抗性基因和HindIII酶切位点。短暂表达的Cre重组酶移除了诺卡因基因座，留下HindIII酶切位点和一个loxP位点。新插入的HindIII酶切位点有助于区分这个等位基因上Tcrb的重排与Tcrbtm1Slek基因座的区别。（来源：J:147947）
