通过以下方式构建了一个表达含有LAT膜和脂筏靶向序列，与全长Lcp2(SSL-76)融合的 chimera分子的等位基因。将小鼠Lat基因的1-105号碱基对与Lcp2的起始exon1和内含子1连接。Lcp2基因的ATG启动子被突变，整个构建位于FRT引导的诺卡因 cassette之后。经过同源重组后，携带正确靶向等位基因的鼠与FLPe转基因鼠杂交，以去除诺卡因 cassette。杂合子胸腺细胞的免疫印迹分析显示了80kDa的融合蛋白和76kDa的野生型蛋白。(来源：J:147721)
