设计了一个靶向载体，目标是将Cre ORF后面跟着一个由FRT座点包夹的PGK-neo cassette插入Tbx18基因座。经过同源重组后，Cre的起始密码子取代了原生Tbx18的ATG。载体经线性化后，通过电穿孔注入129S1/SvImJ ES细胞。通过Southern blot筛选出正确重组的克隆，然后将它们注入CD-1囊胚。产生了杂合雄性，与Cre报告基因Gt(ROSA)26Sortm1Sor的携带者杂交。在E12.5期胚胎中，Cre表达在心外膜、室间隔和左心室心肌壁等多个位置被检测到(引用文献：J:147421, J:160473)。
