The transgene construct contained the cytomegalovirus (CMV) promoter and the beta-galactosidase gene with a nuclear localization signal (nlacZ) separated by a loxP-flanked transcriptional STOP sequence (C-terminal sequence of yeast His3 gene, the SV40 polyadenylation signal (PolyA), and a 5' splice donor site). When mice carrying this transgene are crossed to mice expressing Cre recombinase, the STOP sequence is excised, permitting beta-galactosidase expression. (J:147415)
