使用了一种带有自剪切LoxP位点和S-mu开关区上游的neomycin/sperm特异性Cre cassette的靶向载体,目标对准了LoxP位点和位于E-mu增强子下游的I-SceI内切酶位点。该构建通过电穿孔技术导入了C57BL/6的白化胚胎干细胞(ES)细胞。经过正确筛选的ES克隆被注入受体囊胚中。产生的雄性后代能够自行剪切掉neomycin/Cre cassette,保留LoxP和I-SceI位点在生殖细胞中。通过Southern blotting确认了精确的靶向。杂合子小鼠的B细胞发育和数量正常。(来源:J:145691)
