这个等位基因是通过将一个男性α-βT细胞的核转移到去核的卵细胞中创造出来的。随后培养出一个囊胚，再从中克隆出一个ES细胞。这个ES细胞中的两个Tcra基因，一个发生了有功能的、框内重组，另一个则是无功能的、框外重组。仅推广并描述的这个是含有Valpha3.1-J30重组的有生产性的等位基因。重组性通过Southern blotting和倒置PCR来确定。（J:143006）
