The transgene contains a full length mouse cardiac phospholamban cDNA with two consecutive point mutations (AAT->GCT) that encodes alanine instead of asparagine at amino acid position 27 of the protein (N27A), followed by an SV40 polyadenylation signal, under control of the cardiac-specific alpha myosin heavy chain (myosin, heavy polypeptide 6, cardiac muscle, alpha) promoter. Quantitative immunoblot analysis demonstrated cardiac expression of phospholamban protein in this line (91) at 140 +/- 12% the level observed in wild-type hearts. (J:144921)
