一段5.8千碱基对的rat nestin启动子序列驱动表达一段0.8千碱基对的rat Emx2 cDNA，其序列与mouse Emx2完全相同。随后是SV40的polyA信号，后面跟着5.4千碱基对的nestin基因组序列，提供增强作用。在E14.5时，通过原位杂交可以检测到表达，这是皮层神经生成活动的高峰，特别是在转基因小鼠中同时检测到内源性和转基因表达的Emx2。在转基因小鼠的皮层VZ区域，Emx2转录物呈现出从高背侧到低腹侧的梯度，且表达水平高于对照组。此外，转基因小鼠的神经管旁皮层也显示了增强的Emx2表达，而在nestin阳性增殖区域如dTh，虽然对照组中未检测到，但也能观察到适度的表达。（文献来源：J:96705）
