在XbaI酶切位点，一个LoxP位点替换了3kb的内含子序列，位于第3外显子上游，然后插入了一个被修饰的诺卡因抗性 cassette。通过在正确靶向的ES细胞克隆中产生的小鼠与EIIa-cre转基因小鼠杂交，后代通过Southern blot筛选出丧失诺卡因抗性 cassette的个体。通过免疫印迹检测脾细胞提取物，结果显示蛋白表达没有受到影响。Cre介导的第3外显子切除将去除一个对蛋白质功能必需的调控区域。(来源：J:144598)
