野生型Hesx1的编码序列通过PCR被改造，使其第160位的Arginine变为Cysteine（CGA->TGC）。在靶向载体中，一个含LoxP位点的PGK-Neo选择性 cassette被插入到突变Hesx1序列的首内含子。PGK-Neo的表达方向相对于正常转录方向是反向的，以期降低突变基因在Hesx1基因活跃的ES细胞中可能产生的显性负作用。在CCE ES细胞中，同源重组将野生型序列替换为靶向载体中的突变序列。F1代动物与Actin-beta-cre小鼠杂交，以剔除PGK-Neo cassette。(来源：J:142649)
