野生型Hesx1的编码序列通过PCR被修改，使得第26位（异亮氨酸）的密码子变为编码丝氨酸（Ile→Thr），插入了位于突变序列首内含子的LoxP引导的PGK-Neo筛选 cassette。在靶向载体中，通过同源重组，CCE ES细胞将野生型序列替换为突变序列。F1代动物与Actin-beta-cre小鼠杂交，以去除PGK-Neo cassette。（来源：J:142649）
