将连接在神经调质膜受体palmitoylation位点上的黄色荧光蛋白(YFP-Mem)插入目标基因的首编码区，由Runx3远端启动子驱动表达。在YFP的3'端，包含PGK-诺卡因抗性基因座，其两侧有loxP位点。在CD8+ T细胞和TH1亚型活化CD4+ T细胞中的目标基因蛋白表达几乎不可检测，且前者显著较低。YFP在MHC I限制的胸腺细胞、静息CD8+ T细胞以及活化CD4+ T细胞中表达。(来源：J:141144)
