这个 Rosa-CAG-LSL-tdTomato-WPRE 目标载体的设计如下：从 5' 至 3' 有 CMV-IE 增强子/鸡β-actin/兔β-球蛋白杂交启动子（CAG）、FRT 立即剪切位点、一段 loxP 介导的停泊子（包含所有三种阅读框的终止密码子和三重polyA信号）、tdTomato序列（非聚合的DsRed荧光蛋白变体，通过12个氨基酸连接两个拷贝的蛋白（串联二聚体））、木瓜病毒后转录调控元件（WPRE，以提高mRNA的稳定性）、polyA信号，以及attB/attP引导的PGK-FRT-Neo-polyA插件。整个构建被插入Gt(ROSA)26Sor基因座的外显子1和2之间。通过phiC31重组，Gt(ROSA)26Sortm9(CAG-tdTomato)Hze去除了Neo抗性基因。来源：(J:155793)
