在含有Hras1tm1Xbr的ES细胞中，插入了一个被荧光素标记的终止子（hygro STOP cassette）在编码第1个外显子的上游。同时，第12个密码子的第一个碱基由G变为了T，导致缬氨酸替换为甘氨酸。通过小鼠的生殖细胞（germ line）和Cre介导的重组，去除了这个荧光素标记的终止子。最终得到的突变型携带G12V变异以及插入的IRES-beta-geo序列。（来源：J:137729）
