通过在ROSA位点插入由内源性启动子控制的cre60.F2 N端cre片段，该片段与FRB（FRAP/MTOR的配体结合域，一种FKBP12-rapamycin关联蛋白）相连，并插入一个Caggs-cre59.F2元件，该元件由C末端cre与FKBP12（FK506结合蛋白）连接，受CAG启动子驱动。这样构建的融合蛋白在没有rapamycin刺激时无活性。为了筛选，还插入了hygro选择 cassette。通过F1 Gt(ROSA)26Sortm1(cre)Jphe小鼠与FLPe小鼠杂交，可以去除hygro cassette。来源：(J:130959)
