在基因座位7的外显子7内，通过定向的A到T替换，产生了112和128位的酪氨酸替换成苯丙氨酸的突变。为了靶向操作，我们放置了一个位于7号外显子下游的诺卡因抗性loxP和FRT引导的载体。随后，通过与表达Cre的生殖细胞系杂交，我们移除了选择性载体。通过Western blot对胸腺细胞的分析，这些点突变并未影响基因表达。(来源：J:132995)
