在基因座的第7外显子145位由A替换成T的点突变(Y145F)是通过定向的A到T替换实现的。为了靶向插入，我们放置了一个包含loxP和FRT序列的诺卡因抗性 cassette，位于第7外显子下游。随后，通过暂时的Cre表达，这个选择性 cassette被移除，只留下单个loxP和FRT座。通过 Western blot 对胸腺细胞的分析，结果显示此点突变并未影响基因表达。(来源：J:132995)
