一段带有SV40内含子和poly-A序列的人类RET cDNA，后面跟着带有poly-A和frt-flanked neo cassette的CFP，这些都被插入到了exon 1中，去除了包括起始的甲硫氨酸密码子和前6个氨基酸的信号肽编码序列。通过小鼠的生殖细胞中使用flp介导的重组，neo cassette被移除，留下一个floxed RET和CFP cassette（来源：J:132854）。
