这个构建包含了经过点突变改造，使终止密码子变为BamH1限制酶识别位点的Tcf3基因。增强绿色荧光蛋白（EGFP）的整个编码区域被准确地插入到了这个BamH1位点。下游插入了PGKneo选择性标记元件，同时在外显子区域插入了由PGK驱动的链霉素基因作为阴性选择标记。实验结果显示，小鼠的造血干细胞、骨髓中的B细胞系列（包括B线粒体和红细胞系列）以及外周的B1和B2亚群B细胞都表达出了功能性的Tcf3和EGFP。（来源：J:88032）
