使用两个靶向载体，分别在Snord116基因簇的上游和下游插入了loxP位点（以及Frt1介导的PGK-neo和Frt5介导的 puromycin抗性/TK基因 cassette）。上游载体转染到了C57BL/6衍生的Bruce-4胚胎干细胞（ES细胞），经过筛选，得到正确靶向的ES细胞后，再用下游载体进行转染。双靶向的ES细胞随后短暂转染一个表达Cre的质粒。最终得到的1-loxP ES细胞（Snord116基因簇被删除，保留一个loxP位点）被注入受体囊胚中。（来源：J:131427）
