将包含Neo和TK选择标记的删除 cassette 送入了基因的第二个内含子，并在第四内含子中插入了一个第三个loxP位点。短暂的Cre表达后，筛选出了删除了选择 cassette 但保留了3号和4号外显子由loxP位点环绕的克隆。PCR确认了loxP位点的插入。（来源：J:127458）
