在MEF细胞或转基因乳腺组织中，位于第四外显子上游的Neomycin cassette被插入了floxed突变，而第三个LoxP位点则位于第四外显子下游。这种floxed突变不影响目标基因在纯合子MEF和乳腺组织中的蛋白表达。当与表达Cre重组酶的小鼠杂交后，后代会在Cre表达的组织中缺失第四外显子（编码保守的Sir2结构域）。（来源：J:85542, J:121948）
