A loxP site was inserted upstream of exon 1 and a neo cassette flanked by frt sites and containing a 3' loxP site was inserted downstream of exon 1. The neo cassette and exon 1 was removed by cre-mediated recombination following a cross to Tg(EIIa-cre)C5379Lmgd mice. Insertion and removal of the neo cassette and exon 1 was confirmed by PCR analysis on genomic DNA. Absence of protein was confirmed by western blot analysis on brain, lung and thymus homogenate.
