A targeting vector was designed to replace exons 2-5 and part of exon 6 with a neomycin resistance cassette. Analysis of mutant MEFs determined that a transcript is produced, and sequencing showed it resulted from exon 1 being spliced to exon 7, with a continuous open reading frame. A protein lacking the cysteine-rich domain (exon 2), the FNII domain (exon 3) and the first CTLD (exons 4-6) was expressed. (J:124909)
