在BAC（RPCI23-331K23克隆）中通过同源重组，插入了一个FRT标记的neo cassette（后面跟着loxP位点），它紧接在目标基因的第8外显子下游。当靶向载体从BAC中切割出来时，额外的loxP位点被添加到了第8外显子上游。正确靶向的ES细胞被注入到囊胚中，然后与表达Flpe的 mice杂交以去除neo cassette。来源：(J:124157)
