靶向载体通过定点突变设计，目标替换小鼠IL-4Ra cDNA中1964-65位的Alanine-Guanine对Guanine-Alanine，对应于第576位氨基酸位置的Glutamine替换为Arginine（Q576R）。同时，载体内插入了loxp-标记的PGK-neo cassette在第11个内含子。这个构建被电穿孔注入129X1/SvJ衍生的RW4胚胎干细胞。经过正确靶向的ES细胞短暂转染含Cre表达载体，以去除选择性 cassette（留下一个loxp位点在第11个内含子），然后注射到C57BL/6囊胚中。（来源：J:152376）
