将携带两种突变的murine APP cDNA，Arg609替换为Asp，Lys612替换为Glu，插入到小鼠Thy1基因中，替换掉包含从exon 2到exon 4部分序列的1.5kb片段。构建中去除了内含子3的特异性thymus元素。这个构建通过微注射技术注入了FVB雌性小鼠1.5天胚胎的初级核前细胞。逆转录PCR确认了在杂合子大脑中，转基因APP表达水平大约是内源性App信息的50%。建立了Line 6品系。（来源：J:32213）
