这个转基因是通过将小鼠Prion启动子与BRI-Abeta42融合构建，融合中包含人类类型2跨膜蛋白(BRI或ITM2B)的cDNA序列，与一段"野生型APP695"编码的Abeta42(β42)序列通过类似furin样切割位点相连。BRI的C端23氨基酸被Abeta42序列替换。来自Abeta42血浆水平最高的克隆(BRI-Abeta42A, 12e)的鼠被赠予Jackson实验室。转基因融合蛋白的表达水平接近内源性小鼠APP的表达水平，并且在Prnp启动子特有模式下表达，主要在海马和齿状回的颗粒细胞中看到高表达。全长的Abeta融合蛋白在加工过的β42肽表达（较低水平）下表达得最显著（J:101023, J:120781）。
