An FRT-flanked neo cassette with a single 3' loxP site was inserted between exons 1 and 2 and an additional loxP site was inserted downstream of exon 5. The neo cassette was subsequently removed through flp mediated recombination. In vivo Cre-mediated recombination removed exons 2 through 5 creating a null allele. The loss of transcript was confirmed by RT-PCR. (J:120783)
