设计了一个靶向载体，目标是去除编码在第2和3外显子的bHLHO域。载体中插入了与第2外显子第12个密码子同框的tau-lacZ融合元件，后面跟着一个可删除的诺卡因抗性标记。随后通过Cre介导的重组移除了neo基因。通过RT-PCR确认了转录本缺失，通过原位杂交证实了突变蛋白的表达。(来源：J:119562)
