A linearized bacterial artificial chromosome (BAC) containing the 54-kb human telomerase reverse transcriptase gene, including at least 11 kb of upstream sequence, all of the exons and introns, and 1.2 kb of downstream sequence, was microinjected into C57BL/6 oocytes. Transcript expression patterns of the human gene in most organs of transgenic mice are generally characteristic of TERT expression in humans, rather than of the endogenous mouse gene; the exception is that human TERT, which is not expressed in human brain, is like the mouse enzyme expressed in mouse brain.
