A targeting vector was designed to delete 1.6 kb of genomic sequence covering the promoter, exon 1 and 631 bp of intronic sequence, leaving the Gnas minimum promoter region intact. The vector included a tACE-Cre gene for excision of the cassette in the male germ line. RT-PCR failed to detect transcript in paternally inherited mutants. (J:106793)
