使用了两步同源重组技术，从Tcrb基因座删除了70个碱基对（475kb）。第一步，目标事件替换Dbeta1-Jbeta1簇为一个含loxP标记的neo基因，随后在ES细胞中表达cre，移除neo，留下一个loxP位点。第二次，又在Vbeta1外显子插入了另一个loxP标记的neo基因。随后在选择的ES克隆中通过cre表达，移除了包括5'Vbeta区3'端、基因座中央大内含子以及第一个Dbeta-Jbeta簇在内的大片475kb区域。（J:84291）
