Southern blot analysis showed and interphase fluorescent in situ hybridization (FISH) verified integration on Chr 13 of approximately 6 copies of the 194-kb BAC clone 551D6 from the CITB mouse library, which was made from 129S1/Sv-derived CJ7 embryonic stem cells. This clone contains a 187,2-kb insert that encompassses the entire Nr2e1 gene. Allele-specific RT-PCR of brain mRNA from C57BL/6J mice congenic for the transgene demonstrated expression of the 129S1/Sv-derived transgenic allele at backcross generation N10. (J:105597)
