在转录起始点下游的内含子1位置插入了一个LoxP引导的转录阻断元件(TBC)构建了这个无义等位基因。TBC包含了一个Splice接受器、SV40的polyA信号、SV40增强子、诺卡因抗性基因、两个HSV-TK的polyA序列、一个合成的polyA信号/转录暂停信号，以及另一个合成的polyA序列后面跟着一个与Myc关联的锌指蛋白结合位点。Cre表达会通过去除TBC重新激活这个位点。(来源：J:104704)
