使用了一种靶向载体，替换掉了exon 4、16bp的intron 3内含子和13bp的intron 4内含子，用的是自剪切的tACE-CRE-neo元件。重组导致的框架移位据信会在exon 5处造成提前终止。RT-PCR在大脑、心脏、肾脏、肝脏和肌肉样本中未检测到预期的0.75片段。 Western blot分析这些组织也未发现蛋白质。(来源：J:103742)
