Most of the putative exon 1 containing the first ATG was deleted upon the insertion of an EGFP with a loxP-flanked PGK-neo in reverse orientation. Northern blot and RT-PCR confirmed lack of transcript in mutant liver samples. Absence of protein was demonstrated by Western blot of mutant liver, white adipose tissue and kidney. (J:100209)
