The coding sequence of the transgene consists of the puromycin acetyltransferase coding sequence fused in-frame to a 5' truncated HSV1 thymidine kinase gene that begins at the second ATG of the full-length gene and encodes a protein lacking the carboxy-terminal 45 amino acids. This construct resides upstream of the bovine growth hormone polyadenylation signal and downstream of the phosphoglycerate kinase 1 promoter followed by a loxP-flanked neomycin acetyltransferase gene with the bovine growth hormone poly-A signal.
