将lacZ-neo cassette替换了编码HLH域和C-temrinus基因大部分序列。该 cassette 插入了内切位点，使得能够表达一个融合基因，其编码Id4 N端65个氨基酸与β-半乳糖苷酶的融合蛋白。突变大脑和睾丸的RT-PCR未检测到编码HLH域的mRNA。X-gal染色显示融合基因的表达与内源基因（J:98270）的表达相似。
