在KIX域编码序列的上游插入了floxed neomycin选择 cassette，然后对包含KIX的exon进行了改造，将第650位的酪氨酸替换为异亮氨酸（Y650A），第654位的异亮氨酸替换为谷氨酰胺（A654Q），第658位的酪氨酸替换为异亮氨酸（Y658A）。随后通过短暂的Cre表达，移除了选择 cassette。突变产物的表达和稳定性保持正常。（来源：J:98329）
