构建的靶向载体包含Cre重组酶融合蛋白以及人类孕激素受体修改后的配体结合域，以及pgk-neo-polyA插件，它被插入Grid2基因的起始翻译位点，且保持同源性。大约去除了900bp的内源序列起始部位。孕激素受体的配体结合域被修改，能与合成的激素RU486结合并激活，而不被内源性孕激素激活。在用RU486诱导插入的Cre/PGR表达后，观察到了大脑小脑皮层成熟浦肯野细胞中的活性。(来源：J:97754)
