This transgene uses a Ttr promoter to drive expression of a c-Myc tagged Foxo1, mutated to include a single amino acid substitution replacing the phosphorylation site Ser253 with a non-phosphorylatable amino acid. This line expressed the fusion protein in liver at levels approximately threefold higher than line 307. Expression was also shown in Beta-cell and duct-cells of the pancreas by RT-PCR and immunohistochemistry. Total Foxo1 expression in islets was determined to be fivefold higher in this line than in line 307 or wild-type mice by RT-PCR. (J:79560)
