在loxP位点附近，替换了包含DNA酶I敏感性位点IV（HS IV）的1.9kb区域，HS IV位于Il4和Kif3a基因之间，是一个Il4的抑制子。短暂的Cre重组酶表达在ES细胞中移除了neo cassette。突变细胞的DNA酶敏感性分析显示HS IV缺失，但其他敏感位点未受影响。（来源：J:94039）
