Myh9^{Gt(Betageo-btk)1Dash}

A gene trap vector containing a splice acceptor sequence, promotorless Betageo, pA, PGK, exon 1 from Btk and splice donor sequence, was inserted into intron 1 of the endogenous gene. Btk contained a stop codon in each reading frame to prevent translation of downstream fusion transcripts. Protein expression was reduced by half in heterozygotes as shown by immonoblot analysis.
